Dachaihu Decoction medicated serum inhibiting the AR42J cells inflammatory response by inhibiting the JAK2/STAT3 signaling pathway
Objective To investigate the preventive and therapeutic effects of Dachaihu Decoction(DCH)medicated serum on the inflammatory reaction of pancreatic acinar cells and its possible mechanisms.Methods HPLC was used to detect the contents of index components in Dasaihu Decoction.The acute acinar cell inflammation model was replicated by inducing the AR42J cell with cearulein.Cell viability of different concentrations of DCH-medicated serum was detected by CCK-8.The phosphorylation levels of JAK2,STAT3 and NFκB were detected by Western blot.The contents of IL-6 and TNF-α were detected by ELISA.Results The contents of baicalin and paeoniflorin in Dachaihu Decoction were 13.02 mg/mL and 7.19 mg/mL,respectively.The dosage of DCH-medicated serum was less than 20%.The DCH-medicated serum decreased the contents of TNF-α and IL-6 in the supernatant of inflammatory cells,relieved the phosphorylation of JAK2,STAT3 and NFκB in cells and alleviated the inflammatory response of cells.STAT3 inhibitor cucurbitacin decreased the inflammatory inhibitory effect of Dachaihu Decoction.Conclusion Dachaihu Decoction medicated serum can reduce the acute inflammation in acinous cells by inhibiting the JAK2/STAT3 signaling pathway and further reducing the activity of NFκB,and decreasing the synthesis of pro-inflammatory cytokines.
