Establishment of fingerprints of Rosa rugosa'plena'and their comparison with those of proximate species
Objective To establish the HPLC fingerprints of Rosa rugosa'plena'and compare them with those of Rosa rugosa CV.'plena',Rosa chinensis Jacq.,Rosa gallica,Rosa Jinbian and Rosa crimson glorn.Methods High performance liquid chromatography(HPLC)was performed on the ZORBAX Eclipsc XDB-C18 column(4.6mm×250mm,5µm),with its mobile phase as an acetonitrile(A)-0.1%phosphoric acid solution(B)for gradient elution,with its detection wavelength at 280nm,its column temperature at 35℃,its flow rate at 1.0mL/min,its sample size as 10µL.The fingerprints of 12 batches of Rosa rugosa'plena'and 5 related rose species were evaluated and compared by the Similarity Evaluation System of TCM Chromatographic Fingerprint(2012 edition).Results A total of 10 common peaks were identified,3 of which were gallic acid,rutin and quercetin.The similarity values of 12 batches of Rosa rugosa'plena'were ranging from 0.884 to 0.998,indicating a high degree of similarity.There was a great difference in their fingerprints.Conclusion The fingerprints of Rosa rugosa'plena'can be used as a method for identifying Rosa rugosa'plena'and its proximate species.The method has a strong stability,high precision and good reproducibility,and can reflect the distribution of non-volatile main components in Rosa rugosa'plena'and its proximate species,and can also be used for the overall quality control of Rosa rugosa'plena'.
