Experimental study on the effect of Neiyi Tongjing Ling in the treatment of endometriosis by regulating the Wnt7a/β-catenin signaling pathway
Objective To investigate the effect of Neiyi Tongjing Ling(NYTJL)in the treatment of endometriosis in rats and its molecular mechanism of regulating the Wnt7a/β-catenin signaling pathway.Methods Thirty female SPF SD rats were randomly divided into five groups,with six rats in each.Except for the control group,a model was established in those rats in the sham group,the model group,the high-dose Neiyi Tongjing Ling(NYTJL-H,40 mg/kg)group,and the low-dose Neiyi Tongjing Ling(NYTJL-L,20 mg/kg)group,and corresponding drugs were orally administered in each group.After the intervention,HE staining was used to observe the morphological changes of endometrial tissue.Ki67 immunohistochemistry and TUNEL staining were used to detect the cell proliferation and apoptosis in endometrial tissue.RT-qRT,RT-qPCR and Western blot were used to detect the expressions of Wnt7a and β-catenin in endometrial tissue,and immunofluorescence was used to detect the expression of β-catenin.Results Compared with the model group,the high-dose NYTJL group significantly improved the histological morphology of endometrium,reduced the number of infiltrating cells,promoted cell dispersion,reduced the number of glands,and reduced the volume of glandular cavities.Compared with the model group,both the NYTJL-H group and the NYTJL-L group significantly promoted the apoptosis of ectopic endometrial cells and inhibited their proliferation.Compared with the control and sham groups,the mRNA and protein expressions of Wnt7a and β-catenin in the model group were significantly increased,while compared with the model group,the NYTJL-H and NYTJL-L groups had a significant inhibitory effect on the expressions of Wnt7a and β-catenin.Conclusion NYTJL can inhibit the Wnt7a/β-catenin signaling pathway and promote the apoptosis of ectopic endometrial cells,and thus treat endometriosis.
