长江中游地区水稻中醇溶蛋白的提取与优化

为了提高水稻中醇溶蛋白提取效果,以长江中游水稻为研究对象来优化提取技术.对温度、时间等因素进行控制,通过G-250染色法分析提取效果.实验结果表明,当实验温度为50℃、提取时间为10h、离心环境设置为10000r/min,持续10min、料液比设置为0.025g:1ml、提取液为70%乙醇时,溶酶蛋白提取为最佳条件.同时,与默认条件相比,最佳条件提取效果整体提升7.98%.由此可见,控制好实验条件因素将进一步改善溶酶蛋白提取效果.研究内容将水稻品种鉴定以及蛋白提取提供重要技术支持.
Extraction and Optimization of Alcohol Soluble Proteins from Rice in the Middle Reaches of the Yangtze River
To improve the extraction efficiency of alcohol soluble proteins in rice,the extraction technology was optimized using rice in the middle reaches of the Yangtze River as the research object.Control factors such as temperature and time,and analyze the extraction effect using G-250 staining method.The experimental results show that when the experimental temperature is 50℃,the extraction time is 10 hours,the centrifugal environment is set to 10000 r/min for 10 minutes,the solid-liquid ra-tio is set to 0.025g:1ml,and the extraction solution is 70%ethanol,the extraction of lysozyme protein is the optimal condition.Meanwhile,compared to the default conditions,the overall extraction effect of the optimal conditions is improved by 7.98%.From this,it can be seen that controlling the experimen-tal conditions will further improve the extraction efficiency of lysozyme proteins.The research content provides important technical support for rice variety identification and protein extraction.
