Optimization of Extraction Process and Vitro Antioxidant Activity of Total Polysaccharides from Polygonatum odoratum
The extraction process and antioxidant activity of total polysaccharide from the rhizome of Po-lygonatum odoratum were evaluated.The total polysaccharide was isolated from Polygonatum odoratum rhizome through ultrasound-assisted water bath heating extraction method.The influences of material-liquid ratio,ultrasonic power,ultrasonic extraction temperature on the extraction rate of the total poly-saccharide were investigated.Based on the results of single-factor experiments,the response surface ex-periments give the optimal extraction conditions of total polysaccharide from the rhizome of Polygona-tum odoratum∶material-liquid ratio 1∶15(g∶mL),ultrasonic power 300 W,extraction time 60 min,and ultrasonic extraction temperature 60 ℃.Under these extraction conditions,the total polysaccharide yield of Polygonatum odoratum was 13.58%.The results of the antioxidant activity test showed that when the mass concentration of Polygonatum odoratum polysaccharide was 2.75 g/L,the scavenging rates of Polygonatum odoratum polysaccharide on DPPH radicals and OH radicals were 34.18%and 62.04%,respectively.
