莪术醇抑制脂肪细胞血管生成素样蛋白4逆转卵巢癌细胞卡铂耐药的作用及机制研究

目的:探讨肿瘤微环境中的脂肪细胞介导卵巢癌卡铂耐药的作用机制,并研究莪术醇的逆转作用.方法:CCK-8检测不同浓度的卡铂和莪术醇对人卵巢癌SK-OV-3细胞的毒性作用,选取相应浓度的卡铂和莪术醇进行后续的实验.采用0.4 μm的Transwell小室对脂肪细胞和SK-OV-3细胞进行共培养,克隆形成研究脂肪细胞介导SK-OV-3细胞耐药的作用;采用酶联免疫吸附实验(ELISA)检测条件培养基中的血管生成素样蛋白4(ANGPTL4)的水平;RT-PCR分别检测脂肪细胞和SK-OV-3细胞内ANGPTL4基因的表达水平;Western Blot实验研究脂肪细胞诱导的SK-OV-3细胞内耐药蛋白的表达.结果:卡铂对SK-OV-3细胞的半数抑制浓度(IC50)为52.28 μmol·L-1;莪术醇对SK-OV-3细胞的IC50为91.84 μg·L-1.脂肪细胞与SK-OV-3细胞共培养条件培养基中的ANGPTL4基因的表达水平明显升高(P<0.01),且能够被莪术醇所逆转(P<0.01);莪术醇能够明显抑制共培养条件下脂肪细胞内ANGPTL4基因的表达(P<0.01),对SK-OV-3细胞内的ANGPTL4基因表达不产生明显影响(P>0.05);共培养条件下,SK-OV-3细胞内的耐药蛋白表达明显增加(P<0.01),且这一效应能够被莪术醇所逆转(P<0.01).结论:脂肪细胞与卵巢癌细胞共同培养后能够被诱导活化并分泌ANGPTL4蛋白,促进卵巢癌细胞中耐药蛋白的表达进而介导卡铂耐药,而这一作用能够被莪术醇所逆转.
Effects and Mechanisms of Curcumol in Inhibiting Adipocyte-Induced ANGPTL4-Mediated Resistance to Carboplatin in Ovarian Cancer Cells
Objective:This study aimed to investigate the role and mechanism of adipocytes in carbo-platin resistance of ovarian cancer and explore the reversing effect of curcumol.Methods:The cytotoxic ef-fects of carboplatin and curcumol at different concentrations on SK-OV-3 ovarian cancer cells were deter-mined using the CCK-8 assay.The selected concentrations were then used for subsequent experiments.Co-culture of adipocytes and SK-OV-3 cells was performed using a 0.4 μm Transwell chamber to investigate the role of adipocytes in mediating carboplatin resistance in SK-OV-3 cells.The levels of angiopoietin-like protein 4(ANGPTL4)in the conditioned medium were measured using an enzyme-linked immunosorbent as-say(ELISA).The expression levels of ANGPTL4 mRNA in adipocytes and SK-OV-3 cells were determined by RT-PCR.Western blot analysis was performed to study the expression of resistance proteins induced by adipocytes in SK-OV-3 cells.Results:The half inhibitory concentration(IC50)of carboplatin on SK-OV-3 cells was 52.28 μmol·L-1,while the IC50 of curcumol on SK-OV-3 cells was 91.84 μg·L-1.The expression of ANGPTL4 mRNA in the conditioned medium from the co-culture of adipocytes and SK-OV-3 cells was significantly increased(P<0.01)and could be reversed by curcumol(P<0.01).Curcumol markedly reduced the expression of ANGPTL4 mRNA in adipocytes under co-culture conditions(P<0.01),without significantly affecting the expression in SK-OV-3 cells(P>0.05).The expression of resistance proteins in SK-OV-3 cells was significantly increased under co-culture conditions(P<0.01),and this effect could be reversed by curcumol(P<0.01).Conclusion:Co-culture of adipocytes and ovarian cancer cells can induce activation and secretion of ANGPTL4 protein,promoting resistance protein expression and carboplatin resistance in o-varian cancer cells,which can be reversed by curcumol.
