Distribution of Lactobacillus Plantarum RG-034 Acid-resistant Powders in the Colon Contents of Rats
Objective:To study the effects of acid-resistant embedding technique on colon distribution of Lactobacillus plantarum(L.plantarum)RG-034.Methods:L.plantarum RG-034 acid-resistant powders were prepared by strain culture,fermentation,centrifugation and embedding with acid-resistant protectants.The number of viable bacteria after acid-resistance was determined.Every male Sprague Dawley(SD)rat was i.g.administrated with L.plantarum RG-034 powders(1.0×1011 CFU)or L.plantarum RG-034 acid-resistant powders(3.05×1010 CFU),and the concentrations of L.plantarum RG-034 were measured in the small in-testines and the colons by real time PCR at 6,12,and 24 h after administration.Results:The results showed that the concentration of L.plantarum RG-034 was 6.1×1010 CFU·g-1 in the acid-resistant powders,and the number of live bacteria of L.plantarum RG-034 after acid-tolerance was about 1.4×1010 CFU·g-1.The concentrations of L.plantarum RG-034 increased in the contents of intestinal segments of both the small intestines and the colons in the groups of L.plantarum RG-034 powders compared with those in the control group.The concentrations of L.plantarum RG-034 in the contents of small and colon intestinal seg-ments of rats in the L.plantarum RG-034 acid-resistant powder group were significantly increased.It was further found that L.plantarum RG-034 from the L.plantarum RG-034 powders entered colon after 6 h of administration and subsequently excreted from the body,and the concentrations gradually decreased 24 h after administration.The concentrations of L.plantarum RG-034 for the L.plantarum RG-034 acid-resistant pow-der group in small intestines were higher than those in colons after 6 h of administration,the concentrations in colon increased significantly after 12 h and were higher after 24 h.Conclusion:Compared with the L.plantarum RG-034 powders,the L.plantarum RG-034 acid-resistant powders reaches colons at higher con-centrations,which improves their viability in intestines and provides a basis for clinical applications.
