香旱芹的HPLC指纹图谱研究及含量测定

目的:建立香旱芹HPLC指纹图谱,并测定其中2种特征成分的含量.方法:采用高效液相色谱法,Agilent Eclipse Plus C18(4.6mm×250mm,5μm)色谱柱;流动相为乙腈-0.5%冰乙酸水溶液,梯度洗脱;流速为1.0 mL·min-1;检测波长为321 nm;柱温30℃;进行相似度评价,测定绿原酸、木犀草素含量.结果:10批样品的HPLC指纹图谱中有14个共有峰,绿原酸与木犀草素相似度差异较小,且在进样质量分别在0.1366~0.6829 g和0.2390~1.1952 g范围内与峰面积线性关系良好(r>0.999).结论:建立的香旱芹HPLC指纹图谱能全面反映不同产地的药材质量差异,可用于该品的质量控制.
Quality Control of Cuminum Cyminum by HPLC Fingerprints and Multi-index Determination
Objective:To establish a quality control method for medicine Cuminum cyminum by HPLC fingerprints and determine the content of two characteristic components.Methods:The analysis was devel-oped on an Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)column with the mobile phase comprising of acetonitrile-0.5%glacial acetic acid in water at 1.0 mL·min-1 in a gradient elution and the UV detection at 321 nm.The column temperature was 30℃.Conduct similarity evaluation and determine the content of chlorogenic acid and luteolin.Results:There are 14 common peaks in the HPLC fingerprints of 10 batches of samples,the similarity difference between chlorogenic acid and luteolin was relatively small.The injection amounts of chlorogenic acid and luteolin showed good linear relationship with peak areas within the range of 0.1366-0.6829 μg and 0.2390-1.1952 μg(r>0.999),respectively.Conclusion:The established HPLC fin-gerprint can comprehensively reflect the quality differences of medicinal materials from different origins,and can be used for quality control of the product.
