摘要
目的:建立司美格鲁肽非法添加的专属筛查检测方法.方法:采用Fortis C18色谱柱(2.1 mm × 50 mm,1.7 μm),以0.2%甲酸-乙腈为流动相,超高压液相色谱梯度洗脱分离,ESI正离子化三重四极杆串联质谱法检测.利用色谱保留时间、一级质谱母离子及其二级质谱特征碎片三方面信息,对非法添加的司美格鲁肽进行定性和定量测定.结果:司美格鲁肽质量浓度在25~500ng·mL-1范围内线性关系良好.结论:该方法准确、快速,可应用于司美格鲁肽非法添加的检测.
Abstract
Objective:To establish a specific screening detection method for the illegal addition of semaglutide.Methods:Using a Fortis C18 column(2.1 mm × 50 mm,1.7 μm)and gradient 0.2%formic acid-acetonitrile elution for separation,ultra-high pressure liquid chromatography was performed.ESI positive ionization triple quadrupole tandem mass spectrometry was used for detection.Qualitative and quantitative determination was conducted using information from three aspects:chromatographic retention time,primary mass spectrometry of parent ion,and secondary mass spectrometry of characteristic fragments.Results:Semaglutide was linearly detected in the range of 25-500 ng·mL-1.Conclusion:This method is accurate,fast and exclusive;it can be applied to the detection of illegally added semaglutide.
