Effect and mechanism of miR-557 on human lung cancer low metastatic 95c cells
Objective To investigate the effect and mechanism of miR-557 on human lung cancer low metastatic 95c cells.Methods The miR-557 plasmid(group A)and empty vector(group B)were transfected into 95c cells.The blank cell was taken as the control(group C).The cell proliferation,invasion and migration were detected by MTT assay,Trans well assay and cell scratch assay,respectively.The mRNA and protein expressions of MMP-2 and MMP-9 were detected by real-time fluorescent quantitative PCR and Western blot.Results The inhibition rate of cell proliferation was increased with the extension of time(P<0.05).Compared with group B and group C,the inhibition rate of cell proliferation in group A was higher,the number of transmembrane cells was less,the rate of cell migration was lower and the mRNA and protein expressions of MMP-2 and MMP-9 were lower(P<0.05).Conclusion miR-557 can effectively inhibit the proliferation,invasion and migration of 95c cells.The mechanism for it may be related to the down-regulation of MMP-2 and MMP-9 expressions.
