目的 探讨扶解化瘀颗粒调控线粒体自噬途径对脓毒症小鼠心功能的影响.方法 45只雄性C57BL/6小鼠随机分为五组,每组9只.A、B、M、C组行盲肠结扎穿孔术建立脓毒症模型,S组为假手术组.A组和B组分别于6和12 h后灌胃扶解化瘀颗粒2 mL·mg-1·kg-1,C组6 h后尾静脉注射自噬抑制剂3-甲基腺嘌呤2 mg/kg,M组和S组6 h后尾静脉均注射等量生理盐水.24 h后,检测五组血流动力学指标评估心功能,Western blot法检测五组自噬相关蛋白表达,HE染色观察心肌组织病理学改变,TUNEL染色检测心肌细胞凋亡情况.结果 与S组比较,A、B、C、M组血流动力学指标更差,自噬相关蛋白表达降低,心肌细胞凋亡率均升高(P<0.05).与M组比较,A、B组血流动力学指标改善,自噬相关蛋白表达升高,心肌细胞凋亡率降低,但C组血流动力学指标较差,自噬相关蛋白表达降低,心肌细胞凋亡率升高(P<0.05).A组上述指标改善优于B组(P<0.05).结论 扶解化瘀颗粒调控线粒体自噬途径改善脓毒症小鼠心功能障碍,减轻心肌损伤,减少心肌细胞凋亡.
Effect of Fujie Huayu granules on cardiac function of sepsis mice through regulating mitochondria autophagy pathway
Objective To explore the effect of Fujie Huayu granules on cardiac function of sepsis mice through regulating mitochondria autophagy pathway.Methods Forty-five male C57BL/6 mice were randomly divided into five groups with 9 mice each.The sepsis models were established in groups of A,B,M and C by cecal ligation and perforation.Group S was taken as sham operation group.Group A and group B were given gavage of Fujie Huayu granules 2 mL·mg-1·kg-1 at 6 and 12 hours after model establishment,respectively.The autophagy inhibitor 3-methyladenine 2 mg/kg was injected via the tail vein in group C at 6 hours later,and the same amount of normal saline was injected in groups of M and S at 6 hours later.After 24 hours,the cardiac function was evaluated by hemodynamic indicators,autophagy-related protein expression was detected by Western blot,myocardial histopathological changes were observed by HE staining,and myocardial apoptosis was detected by TUNEL staining in the five groups.Results Compared with group S,the hemodynamic indicators of groups of A,B,C and M were worse,expressions of autophagy-related proteins were decreased,and apoptosis rate of cardiomyocytes was increased(P<0.05).Compared with group M,the hemodynamic indicators were improved,expression of autophagy-related proteins was increased,apoptosis rate of cardiomyocytes was decreased in groups of A and B,but hemodynamic indicators were worse,expression of autophagy-related proteins was decreased and apoptosis rate of cardiomyocytes was increased in group C(P<0.05).The improvements of the above indicators in group A were better than those in group B(P<0.05).Conclusion Fujie Huayu granules may improve cardiac dysfunction,reduce myocardial injury and reduce myocardial cell apoptosis in sepsis mice through regulating mitochondria autophagy pathway.
