REGENERATION AND MONOKARYOTATION OF PROTOPLASTS OF STROPHARIA RUGOSO-ANNULAT
Regeneration and monokaryotation of Stropharia rugoso-annulata protoplasts were investigated. The results showed that the protoplast regeneration was very fast, and the regenerated colonies could be seen by naked eyes after plating 3d. The regeneration rate was 0.97~2.00% on PGPM. The osmotic stabilizers of mannitol, sucrose and magnesium sulphate had no significant effects on regeneration rate, but could affect the morphology of regenerated colonies. Pre-culture of protoplasts in liquid regeneration media for 1~2d at 25oC could decrease the regeneration and monokaryotation rate. The protoplast monokaryotation rate was 77.7%. The regenerated dikaryons and monokaryons could be formed on the plates simultaneously, and had no difference in mycelium growth rate and colony-forming time. Two kinds of parent mating types could be found among protoplasted monokaryons, but the ratio was not 1:1. It was the first report about the regeneration and monokaryotation characters of Strapharia rugoso-annulata.
