Analysis of Small Molecule Substances and their Pathways Promoting the Development of Lanmaoa asiatica Primordium
Lanmaoa asiatica,a rare and wild edible ectomycorrhizal fungus in Yunnan province,has an ability to induce its primordia in the artificial culture medium.In order to understand the key substances and the possible changes of their pathways which can promote the development of primordia in L.asiatica under pure culture conditions,simultaneous use of 1H-NMR,GC-MS and LC-MS metabolomics techniques combined with the application of SMICA-P software was introduced for research in this study.Differential changes of small molecule substances in the primordia and tissue blocks cultured after 35 days were analyzed.As a result,a total number of 370 small molecular substances were identified by performing 1H-NMR,GC-MS and LC-MS detection respectively.Comparative analysis of tissue blocks and primordia revealed the presence of 172 small molecules showing upregulation of its contents,in which 13 substances of fumarate,succinic acid,6-phosphogluconic acid,betaine,glutamine,tryptophan,pyroglutamic acid,trehalose,adenosine,uridine,methanol,ethanol and glycerol detected only in the primordia.These differential small molecules were concentrated mainly in 25 metabolic pathways,among which 10 pathways,such as alanine,aspartic acid and glutamate pathways,were mostly influential.The primary exploration of upregulated differential small molecules and the discovery of enriched metabolic pathways which may play key roles in regulating and promoting the growth and development of primordia reported at the present study,thereafter,should provide a theoretical and important foundation for the artificial cultivation of L.asiatica in future.
