Inhibitory Effect of Ganoderma Fruiting Body Extracts on Lung Metastasis of Melanoma in C57BL/6 Mice
In order to further explore the antitumor activity and its functional mechanism of Ganoderma,a C57BL/6 mouse model of melanoma lung metastasis was established.Drug treatment respectively with the ethanol extracts(EEGL)and polysaccharides(GL-PS)derived from Ganoderma fruiting bodies was performed to the mouse model at doses of 50,100 and 200 mg/kg.After two weeks,lungs of mice in each group were taken out for morphological analysis.Investigation of the pathological changes through HE staining and expression activity of genes related to extracellular matrix degradation and apoptosis in lung tissues through RT-qPCR were also carried out to understand the effects and possible mechanisms of EEGL and GL-PS on metastasis of melanoma in mouse lungs.Our results showed that both of the supply of EEGL and GL-PS could reduce the lung weight and lung index,and the number of metastases in mice as well.The EEGL was found to inhibit the metastasis of melanoma in mouse lungs by up-regulating the gene expression of MMP-2 and MMP-9,and down-regulating the gene expression of TIMP-2.The GL-PS could inhibit the lung metastasis of mouse melanoma,on the other side,by up-regulating the gene expression of Bcl-2 and down-regulating the gene expression of Bax and Caspase-3.The present study thus provides available experiment data for our future research on the bioactivity of Ganoderma and therefore the development of its related products.
