Progress in the Synthesis of Orlistat and Its Intermediates
Orlistat has brought a new approach to the treatment of obesity,and its chemical total synthesis method is the current research hotspot.The total synthesis process of orlistat was reviewed.The synthesis routes of orlistat include lauraldehyde,unsaturated fatty acid methyl ester,1,3,5-cyclohexanetriol and β-ketoester as raw materials,among which lauraldehyde method is the most concerned.Because of its long carbon chain,lauraldehyde can be directly condensed,which greatly shortens the reaction route.Moreover,this method has the advantages of high yield,cheap and easily available raw materials and simple operating conditions,and has been adopted by some domestic manufacturers and has a good prospect.
