正相高效液相色谱法测定尼古丁手性异构体的含量

建立了正相高效液相色谱法测定尼古丁手性异构体R和S含量的分析方法.采用UniChiral CDN色谱柱(4.6 mm×250 mm,5 μm)和紫外检测器进行分析,流动相为90%正己烷-10%乙醇(加入0.1%二乙胺),柱温30℃,波长254 nm.结果表明,尼古丁异构体R和S在10~500 μg/mL范围内线性较好,精密度和回收率能够满足定量要求.方法操作简单,特异性强,结果准确,稳定性好,适用于尼古丁手性异构体的含量测定.
Determination of Nicotine Chiral Isomers by Normal Phase High Performance Liquid Chromatography
A method was establishedfor the determination of chiral nicotine isomers,R and S,using normal phase high performance liquid chromatography(NP-HPLC).UniChiral CDN column(4.6 mm ×250mm,5 μm)and UV detector were used for analysis.The mobile phase consisted of 90%n-hexane and 10%ethanol(with 0.1%diethylamine),the column temperature was 30 ℃,and the detection wavelength was 254 nm.The results indicated that the method exhibited good linearity within a concentration range of 10~500 μg/mL.Precision and recovery rate met the quantitative requirements.The method is simple,highly specific,accurate and stable,thus can be used for the determination of chiral nicotine isomers.
