The Effect of La Element on the Expression of Genes Related to Citric Acid Metabolism in Navel Orange
In order to explore the key light rare earth elements affecting citric acid accumulation in navel orange(Citrus sinensis(L.)Osbeck),and screen the related genes involved in the process of light rare earth elements affecting citric acid accumulation,'Newhall'Navel Oranges were selected as the primary test material to measure the content of light rare earth elements and citric acid during fruit development;Additionally,transcriptome sequencing was performed on'Newhall'navel orange fruits and leaves treated with exogenous light rare earth element solutions of varying concentrations,and sequencing results were analyzed to screen for relevant genes.The results showed that the content of La was significantly correlated with the level of citric acid in the fruit,which was the main element affecting the accumulation of citric acid in the fruit;2 768 differentially expressed genes were obtained through transcriptome analysis,among which five genes involved in citric acid metabolism showed differential expression levels before and after treatment.After treatment with different concentrations of light rare earth element solutions,the expression of CitPEPC1 was up-regulated,and the expression of CitPEPCK1,CitIDH3,and CitGS3 were down regulated in fruits.In summary,the La element hinders the accumulation of citric acid during fruit development by positively regulating the expression of genes related to citric acid degradation and negatively regulating the expression of genes related to citric acid synthesis.
