Effect of different medium on multiplication culture of Acacia melanoxylon
Through improving the basic medium to increase the productivity of No. 21 clone of Acacia melanoxylon which has relatively low proliferation rate, to suit to the large scale production. The optimum concentration of macro elements, 6-BA, IBA and sugar in medium were selected by single factor experiment based on the data of growth and proliferation of shoots, callus quality and biomass of seedlings. The results showed that: In medium, the MS macro elements had significant effect on the callus quality and biomass. The favorable concentrations for growth was without 6-BA, for the multiplication was 0.5 mg/L, in which had 5.4 shoots per plantlet, for the callus yield was 1.0 mg/L. The medium supplemented with 0.5 mg/L IBA was suitable for the growth of shoots. Sugar ranged from 20 to 30 g/L was the best for the growth and proliferation of shoot, callus quality and biomass of the plantlets.
