16S rDNA Sequence Analysis of 24 Strains of Associative Nitrogen Fixation Bacteria from Rhizosphere of Sisal
Through the separation and purification of associative nitrogen fixation bacteria, as well as the determination of their nitrogenase activities by using acetylene reduction method, twenty -four bacterial strains with nitrogen fixation activity were obtained from the rhizosphere of sisal plants, which were planted in the main production area of Hainan, Cuangxi and Guangdong. Their nitrogenase activities varied from 5.8742 to 1765.6590 nmol/( mL · H). The results of sequence analysis of 16S rDNA showed that the amplification section of 16S rDNA from the tested strains had no large changes in the length, and it was about 1500 bp. The 24 strains belonged to 6 different genera, through searching their homologic sequences in the GenBank and constructing their phylogenetic tree.
