一株解淀粉芽孢杆菌的分离鉴定及其生物学特性研究

从土壤中分离筛选到一株具有广谱抗菌活性的拮抗细菌,命名为XZ-1.采用平板对峙培养法测定其抑菌活性,结果显示:XZ-1对大蒜叶枯病菌、草莓炭疽病菌等8种重要农作物病害的病原真菌均具有较强的拮抗活性,且抑菌带直径均大于10mm.采用PCR扩增16SrDNA片段并用MEGA6.0软件构建系统发育进化树,结果表明XZ-1与解淀粉芽孢杆菌16SrDNA序列KY828923聚为一簇,因此初步鉴定该菌株为解淀粉芽孢杆菌(Bacillus amyloliquefaciens).采用单因素试验设计对XZ-1的生物学特性进行研究,结果表明:XZ-1在pH值为6时生长最佳,在氯化钠浓度为0~10g/L之间时生长情况相对较好;对光暗不敏感,致死温度为100℃、10min.
Isolation and Identification of A Strain of Bacillus amyloliquefaciens and Its Biological Characteristics
In this study, we isolated a strain of antagonistic bacterium with broad-spectrum antimicrobial activity from soil, which was named as XZ-1.The antagonistic activity of XZ-1 was tested by using the method of plate confrontation culture, and the results showed that XZ-1 had strong antagonistic activity against eight kinds of plant pathogenic fungi, such as Pleospora herbarum, Colletotrichum fragariae and so on, and the diameter of inhibition zone was all more than 10 mm.The 16S rDNA fragment of XZ-1 was amplified by PCR, its phylogenetic tree was constructed by MEGA 6.0, and this strain was preliminarily identified as Bacillus amyloliquefaciens.Single-factor experimental design was adopted to study the biological characteristics of XZ-1, and the results indicated that: the best pH-value for the growth of XZ-1 was 6, and the suitable salinity for its growth was 0~10 g/L sodium chloride;XZ-1 was not sensitive to light and darkness;the lethal temperature was 100 ℃ for 10 min.
