Isolation and Identification of A Strain of Bacillus amyloliquefaciens and Its Biological Characteristics
In this study, we isolated a strain of antagonistic bacterium with broad-spectrum antimicrobial activity from soil, which was named as XZ-1.The antagonistic activity of XZ-1 was tested by using the method of plate confrontation culture, and the results showed that XZ-1 had strong antagonistic activity against eight kinds of plant pathogenic fungi, such as Pleospora herbarum, Colletotrichum fragariae and so on, and the diameter of inhibition zone was all more than 10 mm.The 16S rDNA fragment of XZ-1 was amplified by PCR, its phylogenetic tree was constructed by MEGA 6.0, and this strain was preliminarily identified as Bacillus amyloliquefaciens.Single-factor experimental design was adopted to study the biological characteristics of XZ-1, and the results indicated that: the best pH-value for the growth of XZ-1 was 6, and the suitable salinity for its growth was 0~10 g/L sodium chloride;XZ-1 was not sensitive to light and darkness;the lethal temperature was 100 ℃ for 10 min.
