Preliminary Study on Hypoglycemic,Hypolipidemic and Anti-aging Activity of Polysaccharides from Hohenbuehelia serotina
This research investigated the in vitro activity of Hohenbuehelia serotina polysaccharides(HSP).HSP was obtained by water extraction and alcohol precipitation,treated with petroleum ether and Sevag and purified by DEAE cellulose-52 ion exchange chromatography and Sephadex G-100 gel chromatography.The inhibition rate of α-amylase and cholesterol esterase and the binding amount of bile salt were investigated.The anti-aging activity of polysaccharides was detected in Caenorhabditis elegans.HSP-Ⅰ,HSP-Ⅱ and HSP-Ⅲ were washed out by pure water,0.1 mol/L NaCl and 0.2 mol/L NaCl respectively.HSP-Ⅰ-1 and HSP-Ⅱ-1 were obtained after further purification by Sephadex G-100 gel chromatography.In the activity test,the inhibition effect on α-amylase of HSP-Ⅰ-1 and HSP-Ⅱ-1 was 1.48%and 2.16%.The highest inhibition rate of cholesterol esterase was 34.26%and 30.24%,and was positively related to the mass concentration of polysaccharides.Compared to polysaccharides from pomegranate peel and Gleditsia sinensis seeds,HSP had great advantage.The average combined amount of sodium cholate,sodium glycholate and sodium taurocholate of HSP-Ⅰ-1 and HSP-Ⅱ-1 was 3.81%and 3.40%(m/m).After treated with 0.5 mg/mL and 1.0 mg/mL polysaccharides,the average life span of C.elegans was increased by 8.6%and 21.6%respectively.There was no reproductive toxicity to C.elegans.The results showed that HSP had potent hypolipidemic and anti-aging activity.This research provided a theoretical foundation for the further development of HSP.
