Optimization of Rooting and Seedling Growth Conditions for Young Sporophytes of Lygodium japonicum
In order to screen the optimal culture conditions for young sporophyte rooting and seedling growth of Lygodium japonicum,and obtain high-quality seedlings,young sporophytes from aseptic germination culture were used as the explants,and the effects of different culture densities(1,3,5 and 7 sporophytes/flask),different concentrations of inorganic salt in culture medium(1/4MS,1/2MS and MS),also sucrose(1%,2%and 3%of concentrations),exogenous hormones NAA and IBA(0.1,0.3 and 0.5 mg/L used singly or in pairs on rooting and seedling growth of young sporophytes were investigated.The results showed that the culture density of 5~7 sporophytes per flask was more suitable,and the young sporophytes grew well.The optimal culture medium for sporophyte rooting and seedling growth was 1/4MS+1%sucrose+0.3 mg/L IBA+0.3 mg/L NAA,which could significantly improve the fresh and dry weight of sporophytes.Transplanting should be carried out after cultivating for about 90 days,while the sporophyte leaves are larger and deeper green,and the root system is good.It should not be too late,otherwise sporophyte would be under nutrition-deficient,and the leaves would turn yellow and wilt.
