Effects of CuSO4 on Acute Toxicity,Liver Antioxidant Enzyme Activity and Tissue Structure of Odontobutis potamophila
In order to explore the acute toxicity effect of copper sulfate(CuSO4)on Odontobutis potamophila,the static biological test method was used to set six concentration gradients of 1.58,2.22,3.16,6.33,12.66 and 15.82 mg/L according to the preliminary experimental results.According to the results of acute toxicity test,four concentration gradients of 0.50,0.75,1.00 and 1.25 mg/L were set for acute exposure test.The activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX)and malondialdehyde(MDA)content in liver tissue and the changes of liver and gill tissue structure were measured at 6,12,24,48,72 and 96 h,respectively.The results showed that the LC50 of CuSO4 to O.potamophila at 24,48,72 and 96 h were 8.38,3.87,2.92 and 2.50 mg/L,respectively,and the safe concentration was 0.25 mg/L.With the increase of time and CuSO4 concentration,the activities of SOD,CAT and GSH-PX increased first and then decreased,and the difference was significant compared with the control group at the peak value.The MDA content showed a trend of decreasing first and then increasing,and the difference was significant compared with the control group at the lowest and peak values.After acute exposure,the liver tissue of O.potamophila showed vacuolization and necrosis,the nucleus dissolved and disappeared,and the gill tissue showed swelling and degeneration,necrosis and shedding.The above results showed that the changes of SOD,CAT,GSH-PX activity,MDA content,and liver and gill tissue structure reflected the degree of damage to the body of O.potamophila,which could be used as a reference for safety risk assessent of O.potamophila breeding in the future.
