Expression of miR-1296,miR-637 and miR-1243 in Different Pathological Types and Stages of Thyroid Cancer
Objective To observe the expression levels of miR-1296,miR-637 and miR-1243 in thyroid cancer(TC)patients with different pathological types and stages,and analyze the relation-ship between miR-1296,miR-637 and miR-1243 and TC patients'pathological types and stages.Methods Clinical data of 100 TC patients admitted to Halexon International Peace Hospital from July 2019 to July 2020 were collected.All patients were subjected to pathological examination,and the patients'cancerous tissues,paracancerous tissues(>2 cm above the tumor border)and nor-mal thyroid tissues were taken.Using reverse transcription polymerase chain reaction(RT-PCR)technology,the levels of miR-1296,miR-1243,and miR-637 in different tissues were detected and analyzed in conjunction with the clinicopathological characteristics of the patients.Results The relative expression levels of miR-1296,miR-637 and miR-1243 in patients with TNM stage Ⅲ-Ⅳ,lymph node metastasis,and pathological type of differentiated type was lower than those in pa-tients with TNM stage Ⅰ-Ⅱ,no lymph node metastasis,and pathological type of medullary carci-noma of thyroid,and undifferentiated type,respectively(P<0.05);the relative expression levels of miR-1296,miR-637 and miR-1243 in cancer tissues was lower than those in paracancerous tis-sues and normal tissues(P<0.05).The results of binary logistic regression analysis showed that high expression of miR-1296,miR-637,and miR-1243 were protective factors for TC patients with pathological type of undifferentiated type and TNM stage Ⅰ-Ⅱ(OR<1,P<0.05).100 TC pa-tients were followed up for 24 months,of which 75 cases(75.00%)had a good prognosis and 25 cases(25.00%)had a poor prognosis;the relative expression levels of miR-1296,miR-637 and miR-1243 in the good prognosis group were higher than those in the bad prognosis group(P<0.05).The results of ROC curves showed that the AUCs of miR-1243 relative expression and combined assay for predicting poor prognosis in TC patients were both>0.70,which had some predictive value,with the combined assay being the highest.Conclusion The relative expression levels of miR-1296,miR-637 and miR-1243 in cancer tissues are closely related to pathological types and stages of TC patients,and all three can be used as molecular markers for determining the pathological type and stage of thyroid cancer,which in turn can guide clinical treatment.
