Effect of Liuwei Dihuang Decoction with Adjuvant Cyclophosphamide on MMP9, NF-kB Expression in H22 Tumor-bearing Mice
Objective:To observe the effect of Liuwei Dihuang decoction(LW) with adjuvant cyclophosphamide(CTX) on MMP9, NF-kB expression in H22 tumor-bearing mice.Methods:40 male Kunming mice were divided into model group, CTX group, LW group and the group using the LW and CTX,each group contained 10 mice.Animal models of H22 tumor-bearing mice was induced with the axillary injection of H22 cells, LW group started medication at molding two weeks before(22g/kg· d), CTX group(50mg/kg · 2d) and the combination group started medication after molding.After 14d modeling, kill the mice and weigh, weighing the tumor tissue and splenic tissue, calculation of the antitumor effect and spleen index, testing the expression of MMP9,NF-kB protein on im-munohistochemistry assay.Results:The CTX group and the combination group have obvious antitumor effects (P<0.01), and the combination group had better effect on antitumor than LW group (P<0.05).LW group Spleen index increased, the difference being statistically significant compared to the model group(P<0.05), LW group and the combination group spleen index all higher than CTX group (P<0.05 or P<0.01).Compared to the model group,the combination group have declined significantly on the expression of MMP9 (P<0.05);LW group, CTX group and the combination group all have declined significantly on the expression of NF-kB (P <0.05 or P <0.01).Conclusion: In this scheme of combination with chemotherapy for hepatocellular carcinoma , can improve theimmune function ,the auxiliary mechanism of LW may not by NF -KB, but by reduced the expression of MMP9 protein in hepatocellularcarcinoma tissue to achieve.
