基于同等药效下2种不同基原凤尾草的质量控制方法研究

目的:通过实验证明2种不同基原凤尾草的药效和质量控制的研究.方法:通过LPS刺激RAW 264.7建立体外细胞炎症模型,采用不同浓度的凤尾蕨与井栏边草提取物进行孵育.采用腹腔注射LPS(50 μg/kg)和D-gal(400 mg/kg)诱导ICR小鼠急性肝损伤,并以灌胃2种药材总提取物进行干预.检测细胞中NO的生成量和动物血清中肝脏酶ALT、AST水平,炎症因子IL-6、TNF-α水平以及肝匀浆中氧化应激因子MDA、SOD活力,观察肝脏组织病理学变化,评价其对LPS/D-gal致小鼠急性肝损伤的保护作用.对凤尾草的性状、显微、薄层色谱进行鉴别,参照2020年版《中国药典》检测方法对其水分、总灰分、酸不溶性灰分、醇溶性浸出物进行测定,利用高效液相色谱法对木犀草素-7-O-芸香糖苷与木犀草素-7-O-葡萄糖苷进行含量测定.结果:确定凤尾蕨与井栏边草的性状特征,根茎叶的横截面与粉末的显微特征,薄层色谱蕨素C的特征.规定凤尾草的水分不得超过10.9%,总灰分不得超过15.0%,酸不溶灰分不超过10.0%,醇溶性浸出物不得少于16.5%.规定凤尾草的质量标志物含量限度,制定木犀草素-7-O-芸香糖苷与木犀草素-7-O-葡萄糖苷的含量测定方法.该方法分离较好,在0.078 2~8.867 8 mg/g与0.112 2~2.997 1 mg/g范围内呈良好的线性关系,平均加样回收率分别为104.02%和92.49%.结论:该方法专属性强,重复性好,可用于凤尾草药材的质量控制.
Study on Quality Control Methods of Two Different Herba Pteridis Multifidae Based on the Same Efficacy
Objective:To prove the efficacy of two different Herba Pteridis Multifidae through efficacy experiments,and the quality control.Methods:The in vitro cellular inflammation model was established by stimulating RAW 264.7 with LPS and incubated with different concentrations of Pteris cretica L.var.nervosa and Pteris multifida Poir extract.Acute liver injury in ICR mice was induced by intraperitoneal injection of LPS(50 μg/kg)and D-gal(400 mg/kg),and was intervened by intragastric administration of the total extracts of the two medicinal materials.The production of NO in cells,the levels of liver enzymes ALT and AST in serum,the levels of inflammatory factors IL-6 and TNF-α and the activities of oxidative stress factors MDA and SOD in liver homogenate were measured,and the histopathological changes of liver were observed to evaluate its protective effect on acute liver injury induced by LPS/D-gal in mice.The character,microscope and TLC were identified,and the moisture,total ash,acid-insoluble ash and alcohol-soluble extract were determined according to the detection method of Chinese Pharmacopoeia(2020 edition).Luteolin-7-O-rutinoside and luteolin-7-O-glucoside were determined by high performance liquid chromatogrhy(HPLC).Results:The characteristics of Pteris cretica L.var.nervosa and Pteris multifida Poir,the cross section of rhizome and leaf and the microscopic characteristics of powder,and the characteristics of thin layer chromatography pterosin C were determined,and it was stipulated that the moisture content of Herba Pteridis Multifidae should not exceed 10.9%,and the total ash content should not exceed 15.0%.The acid-insoluble ash content should not exceed 10.0%,and the alcohol-soluble extract should not be less than 16.5%.The content limit of quality markers of Herba Pteridis Multifidae was determined,and the determination methods of luteolin-7-O-rutinoside and luteolin-7-O-glucoside were established.The method is well separated and has a good linear relationship in the range of 0.078 2-8.867 8 mg/g and 0.112 2-2.997 1 mg/g,and the average recovery is 104.02%and 92.49%,respectively.Conclusion:The method has strong specificity and good repeatability,and can be used for the quality control of Herba Pteridis Multifidae.
