Optimization of Extraction Process of Diosgenin from Smilax Nipponica Based on Response Surface Methodology
Objectvie:To optimize the process of diosgenin in S.nipponica.Ethanol concentration.Methods:Extraction time,solid-liquid ratio,acid concentration,hydrolysis time and hydrolysis temperature were selected as the main factors.The interaction of independent variables and their effects on the yield of diosgenin in S.nipponica were studied by response surface methodology and Box-Behnken design method.The response surface model was established and the optimal solution was obtained.Results:Under the conditions of ethanol concentration 81%,extraction time 80 min,solid-liquid ratio 1∶23,acid concentration 1.27 mol/L,hydrolysis temperature 95℃,extraction time 4.5 h,the actual yield of diosgenin was 1.70 mg/g,which was in good agreement with the prediction value of 1.71 mg/g obtained by response surface fitting equation.Conclusion:The process optimized by response surface methodology is stable and feasible,and can be used for the preparation of diosgenin from S.nipponica.
