Simultaneous Contents Determination of Five Components in Glycyrrhiza by Dual-wavelength HPLC and Principal Component Analysis
Objective:To establish a dual-wavelength HPLC method for simultaneous determination of liquiritin,liquiritigenin,isoliquiritin,isoliquiritigenin and glycyrrhizic acid in glycyrrhiza,providing reference for comprehensive evaluation of the quality of glycyrrhiza.Methods:The HPLC separation was performed on a Zorbax SB-C18 column(250 mm×4.6 mm,5 μm)with gradient elution of 0.1%phosphoric acid in water-acetonitrile,and peaks were detected at 248 nm and 362 nm,the column temperature was maintained at 30℃the flow rate was 1.0 mL/min,The determination results of 15 batches glycyrrhiza uralensis were analyzed by SPSS 21.0 with principal component analysis.Results:The five active ingredients were well separated with good precision,reproducibility,stability,linearity,spiked recoveries and durability.The principal component analysis showed that the overall quality of Xinjiang Glycyrrhiza glabra was good and the differences were small.Conclusion:The method can be used to determine the contents of the five components simultaneously,and it is reliable and simple,and combined with the principal component analysis,it can provide a reference for the comprehensive evaluation of the quality of Glycyrrhiza glabra.
