通脉助孕颗粒的工艺放大及其质量研究

目的:对通脉助孕颗粒的制备进行工艺放大,并建立其质量标准.方法:对通脉助孕颗粒依次进行小试和中试的放大,获得3批中试样品.对中试样品的性状进行规范化描述,采用薄层色谱法(TLC)对通脉助孕颗粒中的陈皮、炒白芍、赤芍和北柴胡进行定性鉴别,采用高效液相色谱法(HPLC)对芍药苷进行定量分析.色谱柱为C18(5 μm,4.6 mm×250 mm),以乙腈-0.1%磷酸(14∶86)为流动相,流速1.0 mL/min,检测波长230 nm,柱温30℃.结果:通脉助孕颗粒的小试、中试工艺验证均符合要求.陈皮、炒白芍、赤芍和北柴胡的薄层色谱主斑点清晰,分离良好,阴性对照无干扰;芍药苷在58.3~583.0 ng范围内线性关系良好,测得平均回收率为99.97%,RSD为0.35%.结论:通脉助孕颗粒的处方工艺稳定可靠,制备的样品性状稳定、质量均一.建立的TLC和HPLC专属性强、重复性好、准确可靠,能够有效评价通脉助孕颗粒的质量.
Process Scale-up of Tongmai Zhuyun Granules and Quality Study
Objective:Process scale-up of the preparation of Tongmai Zhuyun Granules and establishment of their quality standards.Methods:Three batches of Tongmai Zhuyun Granules test samples were obtained by sequentially scaling up the small and medial tests of the pellets.The properties of the medial samples were described in a standardized manner.The qualitative identification of Chenpi,Chaobaishao,Chishao and Beichaihu in the pellets was performed by thin-layer chromatography(TLC).The quantitative analysis of peony glycosides was performed by high performance liquid chromatography(HPLC).The chromatographic column was C18(5 μm,4.6 mm×250 mm)with acetonitrile-0.1%phosphoric acid(14∶86)as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 230 nm with a column temperature of 30℃.Results:The process validation of the small trial and the medial test of Tongmai Zhuyun Granules are in compliance with the requirements.The thin layer chromatography of Chenpi,Chaobaishao,Chishao and Beichaihu showed clear main spots and good separation,with no interference in the negative control.Paeoniflorin showed a good linear relationship in the range of 58.3-583.0 ng;the average recovery was 99.97%(RSD=0.35%).Conclusion:The Tongmai Zhuyun Granules is stable and reliable,and the prepared samples have stable properties and uniform quality.The established TLC and HPLC are extremely specialized,repeatable,accurate,and dependable and can efficiently assess Tongmai Zhuyun Granules.
