Preparation and Release Evaluation in Vitro of pH-time Lag Colon Specific Tablets from Artemisia Rupestris L.
Objective:To prepare pH-time lag colon specific tablets of Artemisia rupestris L.and evaluate release performance in vitro.Methods:Colon specific tablets were prepared by pressing coating and spray coating.The cumulative release of total flavonoids as the index,the prescription proportion of time lag materials for pressing coating,the amount of coating,the proportion of upper and lower fillers,the prescription proportion of pH controlled release layer and the coating weight gain were investigated respectively.Results:After inspection,the optimal technological formula was EC-PEG 6 000(4∶1),0.1 g of pressing coating,upper and lower layers of pressing coating filler(1.5∶1),Eudragit FS 30 D-Eudragit L30 D-55(2∶1),and when the weight gain of pH controlled release layer coating was 7%,the colon specific preparation was not released in 0.1 mol/L HCl(2 h)solution,the cumulative release was less than 10%in pH 6.8 phosphate buffer solution(4 h),and the cumulative release in pH 7.5 phosphate buffer(6 h)was great than 80%.Conclusion:The colon specific tablets can achieve colon specific release and its release is stable in vitro.
