枸杞提取物对过氧化氢诱导的人视网膜色素上皮细胞氧化损伤的保护作用

目的:体外通过过氧化氢(H2O2)诱导视网膜色素上皮(RPE)细胞造成氧化损伤,观察枸杞提取物对人视网膜色素上皮(ARPE-19)细胞活性的影响,探讨枸杞提取物通过抗氧化对ARPE-19 细胞的保护作用.方法:采用不同浓度H2O2诱导体外ARPE-19 细胞的氧化损伤,确定最佳浓度;采用MTT法检测枸杞含药血清对ARPE-19 细胞活力的影响,确定安全有效的药物浓度;观察各组细胞的形态,划痕试验检测各组ARPE-19 细胞的活力;Western Blot法检测各组细胞Nrf2/ARE信号通路相关因子Nrf2 的质蛋白和核蛋白的表达水平;试剂盒检测氧化应激水平的相关指标活性氧(ROS)含量、还原型谷胱甘肽(GSH)含量及过氧化氢酶(CAT)活力.结果:当H2O2 浓度为 200 μmol/L,作用 2 h时,ARPE-19 细胞出现明显的氧化损伤(P<0.01);不同浓度的枸杞含药血清对ARPE-19 细胞的干预效果不同,在 2.5%、5%、10%的浓度下对损伤的ARPE-19 细胞具有明显的促进增殖的作用(P<0.05),故选择 2.5%、5%、10%浓度的枸杞含药血清进行下一步实验,与模型组相比,枸杞含药血清组细胞形态良好,长势均匀;细胞内的ROS含量显著降低(P<0.01),GSH含量明显上升(P<0.05),CAT活力显著升高(P<0.01);Nrf2 质蛋白表达增强(P<0.05),核蛋白表达减弱(P<0.05).结论:枸杞含药血清能够提高ARPE-19 细胞自身的抗氧化能力,从而对ARPE-19 细胞产生保护作用.
Protective Effect of Lycium Barbarum Extract on Hydrogen Peroxide Induced Oxidative Damage in Human Retinal Pigment Epithelial Cells
Objective:To induce oxidative damage to retinal pigment epithelial(RPE)cells by hydrogen peroxide in vitro,observe the effect of lycium barbarum extract on the activity of human retinal pigment epithelial(ARPE-19)cells,and explore the protective effect of lycium barbarum extract on ARPE-19 cells through antioxidant activity.Methods:Different concentrations of hydrogen peroxide(H2O2)were used to induce oxidative damage in ARPE-19 cells in vitro,and the optimal concentration was determined;Using MTT method to detect the effect of medlar containing serum on ARPE-19 cells viability and determine the safe and effective drug concentration;Observe the morphology of cells in each group,and detect the vitality of ARPE-19 cells in each group through scratch experiments;Western Blot method was used to detect the expression levels of Nrf2/ARE signaling pathway related factor Nrf2 protein and nuclear protein in each group of cells;The kit detects the relevant indicators of oxidative stress level,including reactive oxygen species(ROS)content,reduced glutathione(GSH)content,and catalase(CAT)activity.Results:When the H2O2 concentration is 200 μmol/L for 2 hours,RPE-19 cells showed significant oxidative damage(P<0.01);the intervention effect of different concentrations of Chinese wolfberry medicated serum on ARPE-19 cells is different.At concentrations of 2.5%,5%,and 10%,it has a significant promoting effect on the proliferation of damaged ARPE-19 cells(P<0.05).Therefore,2.5%,5%,and 10%concentrations of Chinese wolfberry medicated serum were selected for the next experiment.Compared with the model group,the Chinese wolfberry medicated serum group has good cell morphology,uniform growth;the intracellular ROS content significantly decreased(P<0.01),GSH content significantly increased(P<0.05),and CAT activity significantly increased(P<0.01);Nrf2 protein expression increased(P<0.05),while nuclear protein expression decreased(P<0.05).Conclusion:Medlar containing serum can enhance the antioxidant capacity of ARPE-19 cells,thus having a protective effect on ARPE-19 cells.
