三阴交电温针对早发性卵巢功能不全模型大鼠卵巢储备功能调节的实验研究

目的:观察三阴交电温针对早发性卵巢功能不全(POI)模型大鼠卵巢储备功能的调节作用,为三阴交临床应用提供科学依据.方法:将动情周期规律的 32 只SPF级雌性SD大鼠,随机分为空白组、模型组、三阴交组、非经非穴组,每组 8 只.空白组以 0.9%生理盐水进行灌胃,模型组以雷公藤多苷片混悬液灌胃造模.造模成功次日进行干预,三阴交组取三阴交穴,非经非穴组取非经非穴,2 组均连接温针电针综合治疗仪,选连续波,频率为 2 Hz,每次留针 20 min,每日干预 1 次,连续干预 14 d.空白组和模型组分别在与电温针组相同时辰给予相同时间的抓取、固定,不进行任何治疗.干预结束后各组大鼠在动情间期取材,采用ELISA检测血清促卵泡生成素(FSH)、雌二醇(E2)、抑制素B(INHB)、抗苗勒管激素(AMH)水平;HE染色观察各组卵巢组织形态变化.结果:与空白组比较,模型组卵巢脏器指数,血清E2、AMH、INHB水平均明显降低,血清FSH含量明显升高(P<0.01).与模型组比较,三阴交组卵巢脏器指数,血清AMH、E2、INHB水平均明显升高,血清FSH水平明显降低,差异均具有统计学意义(P<0.05,P<0.01);非经非穴组AMH水平升高(P<0.01).与非经非穴组比较,三阴交组卵巢脏器指数,血清AMH、E2、INHB、FSH水平变化明显,差异均具有统计学意义(P<0.05,P<0.01).结论:(1)三阴交电温针可调节POI模型大鼠性激素水平,改善生殖内分泌环境;(2)三阴交电温针可能是通过调节POI大鼠血清激素、AMH水平,从而发挥改善卵巢储备功能的作用.
Regulation of Electrothermal Acupuncture at Sanyinjiao on Ovarian Reserve Function in Rats with Premature Ovarian Insufficiency
Objective:To observer the regulation of electrothermal acupuncture therapy at Sanyinjiao on ovarian reserve function in rats with premature ovarian insufficiency(POI),provide scientific basis for the clinical application of Sanyinjiao.Methods:Thirty-two SPF female SD rats with regular estrous cycle were selected and randomly divided into blank group,model group,Sanyinjiao electrothermal acupuncture group and non-acupoint electrothermal acupuncture group,with 8 rats in each group(n=8).The blank group was given 0.9%physiological saline by gastric irrigation,and the model group was given Tripterygium glycosides tablet suspension by gastric irrigation.The next day after the end of the model,electrothermal acupuncture intervention was started,Sanyinjiao group took Sanyinjiao points,the non-meridian and non-acupoints were selected in the non-acupoint group.Both groups are connected with the electrothermal acupuncture instrument,continuous wave,frequency was 2 Hz,20 minutes each time,once a day,continuous intervention 14 days.The blank group and the model group were given the same time of capture and fixation as the electric temperature needle group at the same time,without any treatment.After the intervention,samples were collected from each group of rats during the diestrus period,the ELISA method was applied to test the level of follicle-stimulating hormone(FSH),estradiol(E2),anti-mllerian hormone(AMH)and inhibin B(INHB);The morphological changes of ovarian in each group tissue were observed by HE staining.Results:Compared with the blank group,the ovary index,serum E2,AMH,INHB reduced,while serum FSH increased in the model group(P<0.01).Compared with the model group,the ovary index,serum AMH,E2,INHB significantly increased,while serum FSH significantly reduced in the Sanyinjiao group,the differences were statistically significant(P<0.05,P<0.01);serum AMH increased in the non-acupoint group(P<0.01).Compared with the non-acupoint group,the ovary index,serum AMH,E2,INHB,FSH significantly changed in the Sanyinjiao group,the differences were statistically significant(P<0.05,P<0.01).Conclusion:(1)Sanyinjiao electrothermal acupuncture can regulate the levels of sex hormones in POI model rats and improve the reproductive endocrine environment;(2)Sanyinjiao electrothermal acupuncture can improve ovarian reserve function in POI model rats,which is likely to be related with regulating serum hormone and AMH levels.
