Simultaneous Determination of Eleven Flavonoid Components in Astragali Complanati Semen by UPLC-MS/MS
Objective:To establish a method for simultaneous determination of 11 flavonoid components(myricetin-3-O-β-D-glucopyranoside,complanatoside A,complanatoside B,astragalin,calycosin-7-O-β-D-glucoside,rhamnocitrin 3-glucoside,calycosin,isoquercitrin,guaijaverin,formononetin,rhamnocitrin)in Astragali Complanati Semen by ultra high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).Methods:Using Waters ACQUITY UPLC BEH C18 column(100 mm×2.1 mm,1.7 μm),methanol and 0.1%formic acid aqueous solution as mobile phases,gradient elution was performed at a flow rate of 0.2 mL/min.Column temperature was 40℃and injection volume was 1 μL.The positive and negative ion modes and the multi reaction monitoring mode were adopted for the electro spray ionization.Results:Within the concentration range under investigation,there was a good linear relationship(r≥0.999 55)among the 11 flavonoid components;The average sample recovery rate and RSD were ranged from 97.63%-102.87%and 0.96%-2.73%.Conclusion:This method has good accuracy,precision,stability,and linear relationship,and can be used for the determination of eleven flavonoid components in Astragali Complanati Semen.
