Genetic Differentiation of Shiraia bambusicola Populations from East China Revealed by RAPD Analysis
In this research,we have analyzed the genetic diversity of the 32 individuals in 8 populations collected from Jiangsu,Anhui and Zhejiang provinces by employing random amplified polymorphic (RAPD) DNA markers technique in order to clarify the genetic differentiation among different geographical populations of Shiraia bombusicola.We acquired 5 random primers screened from 50 RAPD primers to identify the polymorphism among the tested materials.The results showed that 77 loci were detected,among which 52 were polymorphic loci accounted for 67.53% polymorphism.Cluster analysis by using unweighted pair group method arithmetic average (UPGMA)demonstrated that the 8 populations were mainly divided into three groups,among which population Anji (AJ),population Linan (LA),population Yixing (YX) and population Guangde (GD) were clustered in the same group and the second group included population Ningguo (NG) and population Xiuning (XN),while population Chunan (CA) was the single one of the third group.Genetic analysis demonstrated that population Chunan (CA) had the highest genetic diversity level,while population Anji (AJ) had the lowest.Both Nei's gene diversity index and Shannon's information indexes indicated that the genetic diversity of Shiraia bambusicola was higher at the species level than that at the population level.
