Accelerating Expression of Antidepressant Effect Associate-gene CREB and BDNF of Flavonoid Extract Content from Apocynum venetum Leaves
Apocynum venetum was a traditional drug which was included by Chinese Pharmacopoeia. We extracted the major active component of Apocynum venetum-flavonoid and investigated its antidepressant effect and corresponding mechanisms based on depression animal models. The antidepressant effect and possible mechanism of flavonoid extract from Apocynum venetum leaves (AV-extract) were observed in forced swimming test of mice,administration of AV extract (25 mg/kg, 50 mg/kg and 100 mg/kg, ip) reduced the duration time of immobility significantly (P<0.05) suggested that Apocynum venetum have outstanding effect of antidepression, and the drug action of it is similar with fluoxetine. To further search for the antidepressant mechanism of Apocynum venetum, corticosterone damaged PC12 cell was used as a model, meanwhile Taqman quantitative testing technique was used to test the expression of the brain-derived neurotrophic factor and cAMP response element binding protein of PC12 cells in the presence of corticosterone (0.01mmol/L) and flavonoid extract from Apocynum venetum leaves (25 μg/mL, 50 μg/mL and 100 μg/mL) for 48 h. The result showed that the expression level of BDNF, CREB was reduced significantly after treatmented by corticosterone only, but was elevated significantly by Apocynum venetum treatment which was up-regulated approximately 10 times with dose-dependment corresponding to only corticosterone treated. The result showed that the possible mechanism of Apocynum venetum maybe related to elevation the expression of BDNF, CREB through the signal path AC-cAMP-CREB.
