Primary Study on Mutagenic Effect in Radish by Ethylmethylsulfone (EMS)
Mutated breeding by ethylmethylsulfone (EMS) has several advantages:high mutation rate, much mutated phenotype and low destruction which has been widely applied for crop breeding. In this study, the germination ratio, root length and agronomic traits in field of mutated seeds were investigated. The results showed that germination ratio, root length and the leaf growth of radish were inhibited as the increasing concentration of EMS (from 0.2%to 2%) which indicated the inhibited effect of EMS to radish. Whereas the sensitivity to EMS was different among differ-ent radish cultivators:Duanye-13>Xiakang-40>Chunbai-11-85>Shuanghong No 1. Moreover, the suitable concentra-tion of EMS for radish was 0.4%~0.6%by the selection criteria of lethal concentration 50. The field traits were recorded after soaking seeds of Duanye-13 treated by 0.5%EMS. The abnormities from treated group were found widely which focused on cotyledon curling and tearing, true leaf thicken and epilation by compared with CK group. This study identified the dose of lethal concentration 50 for radish mutation by EMS and confirmed the mutagenic effect of EMS to radish in aspect of seed germination and leaf development which will provide support and refer-ence for radish breeding in future.
