Study on Determination of Anthocyanin from Bletilla striata Flower and Antioxidant Activity in vitro
Bletilla striata is a dry tuber of Orchidaceae Bletilla striata (Thunb.) Reichb.f.and it is an important Chinese herbal medicine. In recent years, the planting area of Bletilla striata is increasing. The flowers of Bletilla striata are generally purple which may be rich in anthocyanins with the biological activity of anthocyanins.Howerer, there was no relevant research report. In this research, the content of anthocyanins in Bletilla striata's flowers was 0.46 mg/g by pH-differential method. The high performance liquid chromatography (HPLC) showed that it contained delphindin (114.528 μg/g),petunidin chloride (22.737 μg/g),pelargonidin chloride (23.736 μg/g),paeonidin (9.030 μg/g) and malvidin (27.831 μg/g).In vitro assay found that the extract ofBletilla striata's flowers had antioxidant activity. The total antioxidant capacity was 173.1 U/mL,the hydroxyl radical scavenging activity was 147.36 U/mL,the DPPH free radical scavenging rate was 85.43% and the superoxide anion scavenging rate was 18.02%.Sensory evaluation showed that the soup color was purple and transparent of Bletilla striata's flowers by soaking in boiling water.Besides,it tasted slightly astringent and sweet after taste with elegant fragrance. The above studies indicated that Bletilla striata's flowers were rich in anthocyanins, with the antioxidant activity and could be developed as beverage.
