Screening of miRNAs Related to Liver Lipid Metabolism in Wuliangshan Black-bone Chicken(Gallus gallus)Based on RNA-seq
In order to study the expression characteristics of miRNA(microRNA)related to lipid metabolism in liver tissue of Wulian-gshan Black-bone Chicken(Gallus gallus)at different developmental stages,liver tissue samples of hens on the shell day(D1)and 168 days of age(D168)were collected as experimental materials.DNBSEQ platform was used for sequencing.DEGseq was used to screen differentially expressed miRNAs and their target genes.Nine differentially expressed miRNAs were randomly selected for RT-qPCR verification.KEGG pathway classification was used to screen out lipid metabolism-related miRNAs and cluster analysis.The lipid metabolism-related miRNA target genes were predicted and GO and KEGG pathway functional enrichment were performed to con-struct a lipid metabolism-related miRNA and target gene association network.The results showed that 106 differentially expressed miR-NAs were screened,including 54 up-regulated and 52 down-regulated miRNAs.41 miRNAs related to lipid metabolism were obtained by clustering.A total of 38 target genes were predicted,and the functional annotation of the target genes was mainly enriched in lipid metabolism-related pathways such as glycerophospholipid metabolism,fatty acid metabolism and sphingolipid metabolism.Novel-gga-miR2311-5p-DGKZ,novel-gga-miR2047-3p-ACACA,novel-gga-miR866-3p-DGKH are candidate miRNA-mRNA interaction pairs re-lated to lipid metabolism.The results suggest that the expression characteristics of miRNA in liver tissue of Wuliangshan Black-bone Chicken are different at different developmental stages.The results provide a reference for the molecular mechanism of miRNA regula-ting lipid metabolism in liver tissue of Wuliangshan Black-bone Chicken.
