Identification of Biomarkers Associated with Gastric Cancer Based on Topologi-cal Analysis Methods
Gastric cancer(GC)is one of the most common types of malignancy.Due to the hidden nature of gastric cancer,early de-tection is difficult,so it is very important to study the biomarkers related to early diagnosis of GC.Three datasets GSE79973,GSE19826 and GSE13911 were downloaded from GEO database and Limma package was used to identify the differentially expressed genes(DEGs).The protein-protein interactions(PPI)network of DEGs was constructed by DEGs,STRING V1 1 database and Cyto-scape.Hub genes were selected through intersection of four topological analysis methods.The hub genes were verified by univariate and multivariate Cox analysis,Lasso regression analysis,survival analysis,pathway analysis and literature method.1 599,333 and 662 DEGs were identified from these three datasets,respectively.Four hub genes(CDK1,AURKA,PTTG1 and UBE2C)were selected through intersection of four topological analysis methods.GO function and KEGG pathway analysis showed that four hub genes were in-volved in extracellular matrix-receptors,AGE-RAGE signaling pathway in diabetic complications,small cell lung cancer and protein di-gestion and absorption pathways.The survival analysis results showed that the four hub genes were significantly correlated with the o-verall survival of GC patients.Among them,the survival rate of CDK1,PTTG1,and UBE2C high expression groups was signifi-cantly lower than that of the low expression group,indicating that the high expression of these three hub genes were associated with poor prognosis in GC patients.Univariate,multivariate Cox analysis and Lasso regression analysis verified that CDK1,UBE2C and PTTG1 might be potential prognostic markers in GC patients.In order to further support the reliability of the results,literature verification method was used to verify that four hub genes were closely related to the occurrence and development of GC from the perspective of biological experiments.Therefore,CDK1,PTTG1 and UBE2C were most likely to serve as potential prog-nostic markers for GC patients.
