Sequencing of the Mitochondrial Genome of Rhopalosiphum maidis and Phyloge-netic Analysis of Aphidini
To investigate the structural features of the mitochondrial genome of Rhopalosiphum maidis and its phylogenetic relationship within Aphidini,high-throughput sequencing technology was utilized to determine the complete sequence of the mitochondrial genome in Rhopalosiphum maidis in this study.In phylogenetic analyses,the mitochondrial genomes of 18 known Aphidini species were selected as the ingroup,while the mitochondrial genomes of 2 Macrosiphini species were selected as the outgroup.The phylogenetic relationships of Aphidini was reconstructed by using maximum likelihood method and Bayesian inference method.The mitochondrial genome of Rho-palosiphum maidis was a circular molecule of 15 096 bp in length,which contained 13 protein-coding genes,22 tRNA genes,2 rRNA genes and a non-coding control region.All 13 protein-coding genes started with the codons of ATN.While 3 protein-coding genes coxl,nad4 and nad5 employed the incomplete T or TA as the stop codon,the remaining 10 protein-coding genes terminated with TAA or TAG.In this research,the phylogenetic relationships among the Aphidini were reconstructed by utilizing mitochondrial genome data,revealing that the phylogenetic trees constructed by the maximum likelihood method and Bayesian inference method are essentially con-gruent.Among them,the genus Melanaphis,Hyalopterus and Aphis were monophyletic groups,while the genus Rhopalosiphum was non-monophyletic group.
