Characteristics and Phylogenetic Analysis of Mitochondrial Genome in Rambu-riella turcomana
Ramburiella turcomana is a common locust in the grasslands of Xinjiang.In this study,the mitochondrial whole genome se-quence of Ramburiella turcomana was obtained based on second generation sequencing technology,and the characteristics were annota-ted and characterized.A phylogenetic tree of Arcypteridae was constructed using neighbor joining method(NJ)and bayesian inference method(BI).The results showed that the Ramburiella turcomana mitochondrial genome was a double-stranded circular DNA,15 670 bp in length,with a clear AT preference(72.21%),encoding a total of 37 genes,including 13 protein-coding genes,22 tRNA-coding genes,2 rRNA-coding genes,and 1 non-coding control region(D-loop region).The Ramburiella turcomana mitochondrial genome un-derwent the DK rearrangement prevalent in Locustodea.The NJ and BI trees constructed on the basis of mitochondrial whole genomes of reported species of Arcypteridae shared the same topology,with Ramburiella turcomana clustered with total 16 species of Arcypterinae locusts and located at the base of the Arcypterinae,and the remaining six species of Ceracrinae locusts clustered in one group,sugges-ting that Ramburiella turcomana was the older species of Arcypterinae.The whole mitochondrial genome of Ramburiella turcomana pub-lished in this study is the first report in Ramburiella and provides a molecular biological reference for the excavation of the whole mito-chondrial genome of other species of Ramburiella.
