Cloning and Analysis of GLDP and Its Promoters from Four Salsoleae Species
Glycine decarboxylase(GDC)is a key enzyme in photorespiration metabolic pathway,and P protein subunit is its decarbo-xylase unit.This subunit is encoded by the small gene family GLDP.GLDP family members are expressed in all photosynthetic cells in C3 plants,but only in bundle sheath cells in C3-C4 intermediate and C4 plants.In order to understand the molecular mechanism of GLDP expression regulation in different photosynthetic types of Salsoleae species,in this study,based on genome-wide data,GLDP and their promoters were cloned and analyzed from the C3 plant Salsola junatovii,C3-C4 intermediate plants Oreosalsola laricifolia and O.ar-busculiformis,C4 plant Xylosalsola arbuscula.The results showed that all four species of GLDP family have only one member,and they have similar gene structure,conserved motifs and domains.GLDP was expressed in roots,stems and leaves of these plants,with the highest expression in leaves,followed by stems and roots.Based on the analysis of GLDP transcriptional expression abundance in the leaves of Salsoleae and other four published representative genera,the transcriptional expression abundance of GLDP gradually decreased in C3-C4 intermediate plants and C4 plants compared with C3 plants.In addition,the GLDP promoter activity of these four plants fused with GUS expression vector was analyzed by using the transient transformation system in the cotyledon of Oreosal-sola laricifolia,and it was found that the GLDP promoter of Salsola junatovii was expressed in all photosynthetic tissues of the co-tyledon.Compared with of 7 d cotyledon,GLDP promoter of Oreosalsola laricifolia,O.arbusculiformis and Xylosalsola arbuscula was specifically expressed only in 14 d cotyledon bundle sheath cells.The search results provided a reference for further study on the tissue specific expression mechanism of GLDP in different photosynthetic types of Salsoleae.
