光甘草定调控NETs抑制细胞焦亡缓解脓毒症肺损伤机制

目的 探讨传统中药光甘草定对脓毒症肺损伤中性粒细胞胞外诱捕网(neutrophil extracellular traps,NETs)形成及细胞焦亡影响.方法 将24只雄性Wistar大鼠,按照随机数字表法分为3组:脓毒症组采用盲肠结扎穿孔术(cecalligation and puncture,CLP)建立脓毒症大鼠模型;光甘草定组(CLP+GLA)行CLP及光甘草定灌胃(30 mg/kg);假手术组(Sham)行盲肠探查术,仅进行盲肠翻动后关腹.12 h后留取血浆、肺泡灌洗液及肺组织标本检测.测定肺泡灌洗液蛋白含量;测定肺组织湿重/干重(W/D)比值;用苏木精-伊红(HE)染色后观察肺组织病理学改变;用酶联免疫吸附实验(ELISA)检测血浆中NETs标志物MPO-DNA复合物水平、焦亡相关炎症因子IL-18、IL-1 β的表达水平;Western blot技术检测大鼠肺组织Caspase-1及Cleaved-caspase-1焦亡蛋白的变化.结果 脓毒症组肺泡灌洗液总蛋白浓度较假手术组明显升高(P<0.01),光甘草定组肺泡灌洗液总蛋白浓度较脓毒症组下降(P<0.05).脓毒症组肺水肿程度明显加重,光甘草定组较脓毒症组肺水肿减轻.光镜下观察肺组织病理结果显示:假手术组小鼠肺组织结构正常,肺泡清晰;脓毒症组肺泡壁广泛增厚,炎性细胞明显浸润;光甘草定组较脓毒症组肺组织损伤明显减轻.ELISA显示:脓毒症组血浆中NETs标志物MPO-DNA复合物水平和炎症因子IL-18、IL-1 β含量较假手术组明显升高(P<0.001),光甘草定组NETs标志物MPO-DNA复合物水平和炎症因子IL-18、IL-1 β炎症因子表达水平较脓毒症组均显著降低(MPO-DNA:P<0.01;IL-18、IL-1 β:P<0.05).Western blot技术结果显示:与假手术组大鼠相比,脓毒症组大鼠肺组织中Caspase-1及Cleaved-caspase-1蛋白阳性信号表达增强;光甘草定组肺组织Caspase-1阳性细胞分布与假手术组相似,Cleaved-caspase-1阳性信号表达高于假手术组.结论 光甘草定通过抑制NETs生成及细胞焦亡,有效减轻肺部炎症,发挥对脓毒症大鼠肺损伤的保护作用.
Glabridine regulates NETs to inhibit pyrodeath and alleviate lung injury in sepsis
Objective To investigate the effect of glabridin on neutrophil extracellular traps(NETs)formation and pyroptosis in rats with sepsis-induced lung injury.Methods Twenty-four male Wistar rats were divided into three groups according to the random number table method.The sepsis group was established by cecal ligation and puncture(CLP).The Glabridin group underwent CLP and glabridin gavage(30 mg/kg)(CLP+GLA).The sham operation group underwent cecal exploration,and only the abdomen was closed after cecal turning(Sham).After 12 hours,plasma、alveolar lavage fluid and lung tissue samples were taken for detection.Then,protein content of the alveolar lavage fluid was determined;The wet/dry weight(W/D)ratio of the lung tissue was determined;The pathological changes in lung tissue were observed after hematoxylin-eosin(HE)staining.The levels of NETs marker MPO-DNA complex and related inflammatory factors IL-18 and IL-1β in plasma were detected by enzyme-linked immunosorbent assay.The changes of Caspase-land Cleaved-caspase-1 protein in lung tissue were detected by Western blot.Results The total protein concentration of alveolar lavage fluid was significantly higher in the sepsis group compared with the Sham group(P<0.01),and it decreased in the glabridin group compared with the sepsis group(P<0.05).Significant aggravation of pulmonary edema in the sepsis group,and the glabridin group reduced pulmonary edema compared with the sepsis group.The pathological results of lung tissue under the light microscope showed:The structure of lung tissue in the Sham group was normal,and the alveoli were clear;In the sepsis group,the alveolar wall was thickened widely and inflammatory cells infiltrated obviously;Compared with the sepsis group,the lung tissue injury was significantly reduced in the light licorice group.The enzyme-linked immunosorbent assay results showed that the levels of NETs marker MPO-DNA complex and inflammatory factors IL-18 and IL-1β in the plasma of the sepsis group were significantly higher than those in the Sham group(P<0.001).The levels of NETs marker MPO-DNA complex and inflammatory factors IL-18 and IL-1β in the glabridin group were significantly lower than those in the sepsis group(MPO-DNA:P<0.01;IL-18、IL-1β:P<0.05).Western blot Technical results showed that the expression of Caspase-1 and Cleaved-caspase-1 protein positive signal was significantly enhanced in the lung tissue of the rats in the sepsis group compared with the Sham group;the distribution of Caspase-1 positive cells in the lung tissue of the sepsis+glabridin group was similar to that of the Sham group,and the expression of Cleaved-caspase-1 positive signal was higher than that of the Sham group.Conclusions Glabridin can effectively reduce lung inflammation and play a protective role in lung injury in septic rats by inhibiting NETs production and pyroptosis.
