Cloning and expression profiling of circadian clock gene HoPer in Holotrichia oblita(Coleoptera:Melolonthidae)
In order to clarify the expression pattern of the period gene(Period,Per)of Holotrichia oblita,this study used RACE to clone the circadian clock gene HoPer from the second instar larvae,and conducted bioinformatics analysis.RT-qPCR was used to detect the expression levels of HoPer in different developmental stages,different tissues of male and female adults and different photoperiods.The results showed that HoPer encoded 1 172 amino acids,with a molecular weight of 129.67 kDa and an isoelectric point of 5.62.The amino acid sequence deduced by HoPer had two PAS and one period C domain,which were typical structural characteristics of insect PER proteins.Expression pattern analysis showed that HoPer was expressed in all developmental stages,but the expression in eggs was significantly higher than that in other developmental stages.The expression level of HoPer in the head and antennae of male and female adults were in the top two,followed by wings and leg,and throax and abdomen were the last two.There was no significant difference in HoPer expression between male and female adults across the five photoperiods.This study provides some reference for further study on the function of HoPer gene and its regulatory role in the biological clock network.
