Microscopic characteristics of female adults and 28S rRNA molecular identification of 9 species of mealybugs in Ficus spp.
In order to identify the species of mealybugs,we were used morphological and molecular biological methods to study the classification and identification of mealybugs in Ficus spp.By making slides of female adultmealybugs,we compared from the antennae,cerarii,disk pores and cylindrical ducts of the main microscopic characteristics,respectively,and made identification key.38 sequences of 28S rRNA gene fragment were amplified,and compared with the GenBank database,the similarity reached 99.35%~100%.The phylogenetic tree was constructed with each mealybug was clustered together with the known mealybug sequences which downloaded from GenBank database separately,and the results of molecular identification were the same as morphological identification.The results showed that 28S rRNA gene fragment identification of mealybugs was feasible,and the method provided scientific basis for monitoring and warning and precise control of mealybugs in agriculture and forestry.
