Isolation,Identification and Antagonistic Screening of Pathogenic Causing Blackleg Disease of Chrysanthemum morifolium
Yunnan is an important flower province in China.Chrysanthemum morifolium,as an advantageous flower product in Yunnan,with the increase of its planting area and planting time,the occurrence of diseases has become more and more serious,which greatly limits the development of chrysanthemum industry.In 2021,during the investigation of Kunming Flower Demonstration Park in Yunnan Province,we found a disease affecting the base of chrysanthemum stems,which can lead to browning of chrysanthemum stems at the early stage of the disease,and vascular bundles become hollow,and then the whole plant will be atrophied and necrotic at the later stage.In this paper,the pathogen was isolated by tissue isolation method after collecting diseased plants.According to Koch's rule,the pathogen was identified as Dickeya dianthicola based on 16S rRNA and recA gene analysis.27 prokaryotic microorganisms isolated in the laboratory were used as the experimental strains,and the antagonistic bacteria were screened by the plate diffuse method,and 1 strain with inhibitory effect was screened,which was identified as Providencia entomophila by 16S rRNA gene analysis.This study identified the pathogen of chrysanthemum blackleg disease in Kunming City,and provided strain resource for the biological control of chrysanthemum blackleg disease caused by Dickeya dianthicola.
