Objective To investigate the changes of gray matter volume and functional connectivity of some brain re-gions in patients with Diabetic retinopathy(DR).Methods A total of 20 DR individuals(DR Group,n=20)and 23 healthy controls(HCs,n=23)were enrolled in this study.We performed psychological cognitive testing,structural and rest-ing functional magnetic resonance imaging(fMRI)scans respectively.Voxel-based morphometry(VBM)was used to ana-lyze gray matter volume,and the regions with volumetric variation were selected as regions of interest(ROI)for whole-brain functional connectivity(FC)analysis.The gray matter volume and functional connection were compared by two-sample t test.spearman correlation analysis was used to investigate the relationship between gray matter volume(GMV)and func-tional connection values in different brain areas and psychological cognitive test results.Results Compared with the HC group,the gray matter volume of the right medial frontal gyrus(MNI:x,y,z=40.5,10.5,42)and the right fusiform gyrus(MNI:x,y,z=30,-49.5,-12)decreased in DR Patients.Gray matter volume increased in the right orbital medial fron-tal gyrus(MNI:x,y,z=9,67.5,-7.5),right thalamus(MNI:x,y,z=10.5,-9,3),left medial occipital gyrus(MNI:x,y,z=-40.5,-85.5,24),right precental gyrus(MNI:x,y,z=54,1.5,46.5).With the right precental gyrus as the seed point,functional connections with the left superior temporal gyrus(MNI:x,y,z=-60,-30,18)and the left median cin-gulate and paracingulate gyri(MNI:x,y,z=-6,0,33)were weakened.There was no significant correlation between GMV and FC values and psychological cognitive outcomes in DR Patients.Conclusion In DR patients,the volume of gray mat-ter in some areas of the brain is changed,and functional connections with multiple brain regions are impaired,which may be one of the neuropathological bases of cognitive,emotional and behavioral disorders.
