LncRNA DANCR participates in osteogenic differentiation and migration of periodontal ligament cells through negative regulation of miR-216a expression
Objective:To explore the effect of long non-coding RNA differentiation antagonizing non-protein coding RNA(lncRNA DANCR)on the osteogenic differentiation and migration of human periodontal ligament stem cells(hPDLSCs)and its underlying mechanism.Methods:The protein expression of osteogenic markers alkaline phosphatase(ALP),osteocal-cin(OCN)and Runt-related transcription factor 2(RUNX2)was detected by Western blot.The levels of miR-216a-5p and DANCR were detected by qRT-PCR.The relationship between miR-216a-5p and DANCR was assessed by luciferase reporter gene,RNA immunoprecipitation.Transwell assay was used to detect cell migration ability.Results:The levels of DANCR ex-pression decreased and miR-216a-5p expression increased after PDLSCs were stimulated by osteogenic differentiation induc-tion medium.DANCR overexpression or miR-216a-5p knockdown both reduced the protein expression of ALP,OCN and RUNX2 in PDLSCs,and inhibited ALP activity and cell migration ability.There was a targeting relationship between DANCR and miR-216a-5p.Conclusion:DANCR may participate in the osteogenic differentiation and migration of PDLSCs by regula-ting the expression of miR-216a.
