Study on the relationship between pyroptosis and primary Sj?gren's syndrome
Objective:To explore the relationship between pyroptosis and primary Sjögren's syndrome.Methods:A253 cell line was treated with IFN-γ.Cell viability was detected by CCK-8 assay and cell viability staining.mRNA levels of pyroptosis-related markers were measured by quantitative real-time PCR(qRT-PCR).Protein levels of pyroptosis-related markers were detected by immunofluorescence.Immunohistochemistry was performed to detect the protein levels of pyroptosis-related markers in human tissue samples.A253 cells treated with IFN-γ and DMSO were separately treated with the pyropto-sis inhibitor Vx765.Cell viability and mRNA levels of related factors were detected in corresponding groups.Results:The via-bility of cells in the IFN-γ treatment group decreased significantly.mRNA and protein levels of pyroptosis-related markers in-creased.The expression of pyroptosis-related pathway proteins in human tissue samples also increased.Furthermore,after treatment with Vx765,the viability of cells in the IFN-γ group increased,and mRNA levels of pyroptosis-related inflammatory factors decreased.Conclusion:Primary Sjögren's syndrome may be accompanied by pyroptosis during its occurrence.
