目的:深入探讨SPINK5在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达水平与其临床发展和预后的关系,分析其潜在的机制和临床价值,评估其作为靶向治疗基因的潜力.方法:表达量及临床病理数据来自于TCGA数据库中筛选和获取口腔组织来源的癌症病例样本.分析OSCC临床病理特征及预后和SPINK5的相关性.SPINK5在OSCC组织中的差异表达通过免疫组化进行验证.采用基因功能富集分析(gene set enrich-ment analysis,GSEA)方法预测SPINK5调控的信号通路.结果:SPINK5表达量与OSCC临床预后具有显著相关性(P<0.05).高表达组生存时间较长.SPINK5在正常黏膜组织中的表达量均高于OSCC,且差异有统计学意义(P<0.05).SPINK5差异表达的基因富集到了化学刺激的感官知觉、RNA的基因沉默、RNA介导的翻译抑制导致的基因沉默、气味的感官知觉4个生物学过程,和嗅觉受体活动、转录后基因沉默中涉及的RNA结合、mRNA的3'UTR、嗅觉结合4个分子功能.结论:在OSCC中,SPINK5的表达量相对于正常黏膜组织下调,其表达变化与OSCC的预后密切相关.其可能通过参与和调控OSCC细胞的RNA转录和翻译过程来干预OSCC的发生发展.故SPINK5可能可以作为预测患者预后的有效生物标志物和靶向治疗的靶向基因.
Expression of SPINK5 in oral squamous cell carcinoma and its influence on prognosis
Objective:To deeply explore the relationship between the expression level of SPINK5 in oral squamous cell carcinoma(OSCC)and its clinical development and prognosis,analyze its potential mechanisms and clinical value,and e-valuate its potential as a targeted therapy gene.Methods:The expression levels and clinical pathological data were screened and obtained from the TCGA database for oral cancer case samples from tissue sources.The correlation between the clinical pathological characteristics and prognosis of OSCC and SPINK5 was analyzed.The differential expression of SPINK5 in OSCC tissues was verified by immunohistochemistry.The signaling pathways regulated by SPINK5 were predicted using gene set en-richment analysis(GSEA)method.Results:The expression level of SPINK5 was significantly correlated with the clinical prog-nosis of OSCC(P<0.05).Patients with high expression had longer survival times.The expression level of SPINK5 in normal mucosal tissue was higher than that in OSCC,and the difference was statistically significant(P<0.05).The differentially ex-pressed genes enriched by SPINK5 were involved in four biological processes:chemical stimulation sensory perception,RNA gene silencing,RNA-mediated translational inhibition leading to gene silencing,and olfactory sensory perception;and four mo-lecular functions:olfactory receptor activity,RNA binding involved in transcriptional gene silencing,mRNA 3'UTR,and ol-factory binding.Conclusion:In OSCC,the expression of SPINK5 is downregulated compared to normal mucosal tissue,and its expression changes are closely related to the prognosis of OSCC.It may interfere with the occurrence and development of OSCC by participating in and regulating the RNA transcription and translation processes of OSCC cells.Therefore,SPINK5 may serve as an effective biomarker for predicting patient prognosis and a targeted therapy gene.
